Abstract:

Nanotechnology promises enhanced drug efficacy of drugs already being used in the clinic, exemplified by our nanoparticle-loaded epigenetic modulators, improving specificity and tolerability. We developed fabrication methods with a translation focus employing our proprietary nanoprecipitation method. Despite promising pre-clinical data, there have been significant nanoparticle failures in clinical trials highlighting challenges in transitioning to clinical success. A limited understanding of nanoparticle structures and their biological interactions contributes to these failures. Understanding the intricate interplay between nanoparticle properties and their biological interactions is crucial. We report structural properties of nanoparticles using characterization techniques including Cryo-EM, powder-X-Ray Diffraction, Raman, and X-ray Photoelectron Spectroscopy inform the size, morphology, surface chemistry, packing and crystallinity which dictate the breadth of structural-biological interactions and thus nanoparticle fate in vivo. Thus, insights into nanoparticle structure offer untold/undiscovered relevance in translational research.
